The osteogenic differentiation of human dental pulp stem cells in alginate-gelatin/Nano-hydroxyapatite microcapsules

Abstract Background Microcapsule is considered as a promising 3D microenvironment for Bone Tissue Engineering (BTE) applications. Microencapsulation of cells in an appropriate scaffold not only protected the against excess stress but also promoted cell proliferation and differentiation. Through current study, we aimed to microcapsulate human Dental Pulp Stem Cells (hDPSCs) evaluated osteogenic differentiation those by using MTT assay, qRT-PCR, Alkaline phosphatase, Alizarine Red S. Results The SEM results revealed that Alg/Gel microcapsules containing nHA showed rough more compact surface morphology comparison with microcapsules. Moreover, microencapsulation Alg/Gel/nHA could improve induce cultured 1.4-fold 1.7-fold activity BMP-2 gene expression control group after 21 days. mentioned amounts were 2.5-fold 4-fold 28 nHA, addition hDPSCs-laden microcapsule, up-regulate bone-related expressions osteocalcin, osteonectin, RUNX-2 during days through culturing period, too.
